GENOMICS!OF!DIFFUSE!LARGE!B!CELL!LYMPHOMA!
Diffuse! Large! B! Cell! Lymphoma! (DLBCL)! represents! the! most! common! form! of! B! cell!
lymphoma,! accounting! for! ~30%! of! the! de>novo! diagnoses! and! also! arising! as! a! frequent!
clinical! evolution! of! Follicular! Lymphoma! (FL)! and! Chronic! Lymphocytic! Leukemia! (CLL).!
DLBCL!derive!from!the!neoplastic!transformation!of!B!cells!within!the!germinal!center!(GC),!
the!site!of!B!cell!antigen!affinity!maturation,!somatic!hypermutation!(SHM)!and!class!switch!
recombination! (CSR)! of! the! immunoglobulin! genes.! ! These! tumors! include! phenotypic!
subtypes! of! biological! and! clinical! relevance,! including! germinal! center! B! cell>like! (GCB)>
DLBCL,! deriving! from! GC! light>zone! (LZ)! B>cells,! and! activated! B! cell>like! (ABC)>DLBCL,!
originating! from! a! later! stage! of! GC! differentiation! when! B! cells! are! committed! to!
plasmablastic!differentiation.!Structural!and!functional!analysis!of!the!DLBCL!coding!genome!
has! identified! genetic! lesions! that! recurrently! affect! key! functions! of! the! GC,! and! are!
preferentially!clustered!in!distinct!genetic!subgroups!of!the!disease.!Recent!advances!will!be!
presented! in! two! related! areas:! i)! identification! of! GC! subpopulations! based! on! single>cell!
analysis!of!normal!human!GC!B!cells,!which!led!to!further!definition!of!the!cell>of!origin!of!
DLBCL!and!the!identification!of!novel!prognostically!significant!subgroups;!ii)!genome>wide!
structural!and!functional!analysis!of!the!non>coding!genome,!which!led!to!the!identification!
of!a!pervasive!mutational!mechanism!affecting!non>coding!regulatory!regions.!
